Tuna are highly priced fishes that are often used in processed products. For effective fishery management and protection of consumers’ rights, it is important to develop a molecular method to identify the species of the tuna products. In this study we have developed a molecular method based on real-time polymerase chain reaction (real-time PCR) technology for the rapid identification of four tuna species. Four speciesspecific TaqMan probes were designed to identify bigeye tuna (Thunnus obesus), Pacific bluefin tuna (Thunnus orientalis), southern bluefin tuna (Thunnus maccoyii), and yellowfin tuna (Thunnus albacares). A SYBR green system was also designed to enhance the authentication of T. obesus. Both systems can distinguish target species from others in an efficient and high-throughput manner and can be applied to species identification of tuna products. 2012 Elsevier Ltd. Development of New Modified Simple Polymerase Chain Reaction and Real-time Polymerase Chain Reaction for the Identification of Iranian Brucella abortus Strains
Brucellosis is primarily a worldwide zoonotic disease caused by Brucella species. The genus Brucella contains highly infectious species that are classified as biological threat agents. In this regard, the identification of Brucella can be a time-consuming and labor-intensive process posing a real risk of laboratory-acquired infection to the laboratory staff. This stud...
متن کاملDetection of Nocardia Asteroides Complex in Clinical Isolates by Real-Time Polymerase Chain Reaction
Background and Aims: Nocardia asteroides complex is the most common cause of infectious diseases due to nocardiosis. Interspecies differentiation of Nocardia genera is essential for prognosis and timely proper treatment, as well as for epidemiological studies. Since each genus has its own antibiotic resistance, precise careful diagnosis is of prime importance. As compared to biochemical and phe...
متن کاملPnm-15: IUD Insertion and Alterations of Candida Species in Cervicovaginal Specimens by Polymerase Chain Reaction Technique
Background: Vulvovaginal Candidiasis is one of the most common vaginal infections and identification of candida species can be helpful in medical treatment. Intra uterine devices (IUDs) are effective, long-term methods of contraception, which in some cases may lead to Vulvovaginal Candidiasis. This study was developed to assess the effect of IUD insertion on Candida fungal flora in cervicovagin...
متن کاملTaqMan real-time PCR: a reliable method to detect meat species
The aim of this study was to detect and identify meat species (beef, lamb, pork, chicken, donkey, and horse) in Kebab as a Halal meat-based food sold in on-road-restaurants of North Khorasan province (Iran) using TaqMan real-time polymerase chain reaction (PCR). Raw Kebab samples (150 samples) were obtained from on-road restaurants. The samples were prepared, DNA was extracted and TaqMan real-t...
متن کاملDevelopment and Evaluation of Real-Time Reverse Transcription Polymerase Chain Reaction Test for Quantitative and Qualitative Recognition of H5 Subtype of Avian Influenza Viruses
Avian influenza viruses (AIV) affect a wide range of birds and mammals, cause severe economic damage to the poultry industry, and pose a serious threat to humans. Highly pathogenic avian influenza viruses (HPAI) H5N1 were first identified in Southeast Asia in 1996 and spread to four continents over the following years. The viruses have caused high mortality in chickens and various bird species ...
